چکیده

T HE RATIO OF CARBON MONOXIDE ( CO) to oxygen bound by hemoglobin A is 217 times greater than the ratio of these gas tensions in the equilibrium medium.’ This report shows that hemoglobin F has less preferential binding of CO over 02 than hemoglobin A. Because of this difference between adult and fetal hemoglobin the diffusion gradient for CO across the placenta can not be equated with the difference in carboxyhemoglobin per cent saturation ( COHb ) . This relationship is of interest when comparisons are made between maternal and fetal COHb levels to determine the direction of CO transfer. Such comparisons are pertinent when CO is used to evaluate placental function,2 and when CO is implicated as mcdiating an adverse effect on the fetus from maternal smoking.3’4

Oxygen and carbon monoxide equilibria of human adult hemoglobin at atmospheric and elevated pressure.

By F. LEE RODKEY, JOHN D. O’NEAL, AND HAROLD A. COLLISON T HE FUNDAMENTAL LAW describing the competitive combination of oxygen and carbon monoxide with hemoglobin was defined by Douglas, Haldane, and Haldane’ in 1912. At equilibrium the concentration of these two gases 1)ound to hemoglobin and in the gaseous phase were related by the equation : ( COHb) X ( 02)/( O2Hb) X ( CO ) K where K is the ...

Change in Oxygen Absorption of Human Adult and Fetal Hemoglobin Due to 940 MHz Electromagnetic Field Radiation Exposure

The effects of electromagnetic fields (EMFs) radiation at the frequency of 940 MHz on the structure and function of human adult and fetal hemoglobin (HbA and HbF) were studied. After extraction and purification of HbA and HbF, the oxygen absorption values for exposed and unexposed HbA and HbF to EMF were compared. The slope of oxygen absorption curve for exposed HbA was increased while that for...

New Sequential Model for Human Hemoglobin: Alpha Subunit as Cooperativity Inducer

Hemoglobin is a tetrameric oxygen transport protein in animal bodies. However, there is a paucity of information regarding differences between alpha and beta subunits of hemoglobin in terms of oxygen affinity. The sequential model of Koshland, Nemthy and Filmer (KNF model) has attributed similar affinities to both alpha and beta subunits. The main purpose of the present study is to construct a ...

Reliability of the determination of whole-blood oxygen affinity by means of blood-gas analyzers and multi-wavelength oximeters.

Determination of the oxygen affinity of human whole blood with the help of blood-gas analyzers and multi-wavelength oximeters is compared with an accurate method for recording hemoglobin oxygen dissociation curves (Clin Chem 1982;28:1287-92). P50 (oxygen tension at half saturation; So2 = 50%) and Hill's n (delta log [So2/(1-So2)]/delta log Po2) were determined in blood of 24 healthy donors. Thr...

Effect of carbon dioxide on human hemoglobin. Kinetic basis for the reduced oxygen affinity.

The kinetic basis for the reduction in the oxygen affinity of hemoglobin due to carbon dioxide at constant pH, is studied by measuring the deoxygenation rates of oxyhemoglobin with dithionite, as well as the rate of binding of carbon monoxide to deoxyhemoglobin. These studies were performed in buffer containing 0.05 M potassium phosphate, as well as various mixtures of potassium chloride and po...
